The episode is titled “O Brother, Where Art Thou?” and this will be the episode that introduces Jon Cryer as the well-known Superman villain. In the Supergirl universe, Luthor has already been sent to prison by Superman, but shows up in National City due to his release, which is apparently, the result of poor health. While in National City, he will look to recruit his sister Lena to help cure him.
There’s no specification as to what is making him so ill, but if the show follows a common storyline, then it’s a prolonged exposure to Kryptonite that is killing Lena. Luthor is known for wearing a Kryptonite ring to keep Superman at bay, but this tactic is the very thing that is weakening him to the point of killing him.

Secretly furloughed from prison due to failing health, Lex Luthor (guest star Jon Cryer) visits his sister, Lena (Katie McGrath), to seek her help finding a cure. Savvy as ever, Lena is suspicious of Lex’s motives but when she’s faced with a life and death situation, she must decide how she truly feels about her brother.
Meanwhile, Supergirl (Melissa Benoist) and J’onn (David Harewood) face off against Manchester Black (guest star David Ajala). James’ sister Kelly (guest star Azie Tesfai) comes to town.
